                 Diagnosis and Treatment of Cardiac Diseases with the Assistance of Electrical and Biomedical Engineering

报告主题简介:Atrial fibrillation (AF) and post-operative AF is the most common sustained cardiac arrhythmia. Current therapies are suboptimal with limited efficacy and significant toxicity. Gene therapy becomes an attractive alternative to prevent AF. AF is considered to be sustained by reentrant mechanisms that are promoted by atrial remodeling. We studied the two major forms of atrial remodeling: structural and electrical remodeling.Increased Ca2+/calmodulin-dependent protein kinase II (CaMKII) activity occurs in persistent AF. We evaluated the objective in a porcine AF–heart failure model using atrial gene transfer. Inhibiting CaMKII preserved atrial contractile function and attenuated atrial hypertrophy, fibrosis, and apoptosis. We also examined the preclinical efficacy and safety of Potassium voltage-gated channelKCNH2-G628S gene transfer to prevent post-operative AF.Ischemic heart disease remains a major healthcare issue. Heart attack (myocardial infarction) leads to permanent scarring and adverse remodeling. A reliable, cost-effective method for quantification of left ventricular regional function for localization of the infarction zone would be valuable. 4-dimensional echocardiography (4DE) for strain analysis is ideally suitable for this task.Speckle tracking, shape tracking and machine/deeplearning-based image analysis methods have been developed and used on synthetic and canine/porcineinfarction model.Hydrogels will be delivered to the infarction zone to improve the ventricular mechanical function with the guidance of strain analysis.
报告人简介:刘钊,2007年本科毕业于澳门太阳城赌城光信息科学与技术专业。本科期间参与空间无线激光通信研究。2009年,获得纽约州立大学(State University of New York at Binghamton)电子工程硕士。2009-2016,在凯斯西储大学(Case Western Reserve University)和麻省医学院(University of Massachusetts medical school)博士研究,2017年取得凯斯西储生物医学工程博士,主要从事基因治疗心律不齐(心房颤动)的研究。2017年至今,在耶鲁大学做博士后研究,主要研究三维心脏超声的图像处理(斑点追踪,机器学习),用于心肌梗塞的诊断,并使用高分子材料治疗。在国际同行评审的医学领域杂志: JTCVS, Heart Rhythm, AER, JACC basic to translational science, 以及Nature发表多篇论文。
